Freezing pollen sacs from a sts treated female

Can you freeze pollen sacks from a sts treated female and use them at a later date. Say freeze them just as there about to release pollen ?

A storage method I’ve heard is mixing 1 to 10 pollen to baked dry flour. Then in a sealed jar with silica packets.

FYI strength of the sts concentration makes a direct impact on the number of mature pollen cells.

3m is the recommended concentration. Here’s a recipe for 8oz

A) 3 mM STS (recommended)

Target [Ag] = 0.003 mol/L.

Moles Ag needed: 0.003 × 0.2366 = 0.0007098 mol.

Weigh:

AgNO₃: 0.0007098 mol × 169.873 g/mol = 0.121 g.

Na₂S₂O₃·5H₂O: (2 × 0.0007098) mol × 248.18 g/mol = 0.352 g.

Procedure:

  1. Dissolve 0.352 g sodium thiosulfate pentahydrate in ~150 mL deionized water.

  2. In a separate amber container, dissolve 0.121 g silver nitrate in ~20–30 mL water.

  3. With rapid stirring, slowly add the AgNO₃ solution into the thiosulfate solution (never the reverse). The complex forms immediately; solution should remain clear.

  4. Rinse the AgNO₃ vessel with a bit of water into the mix, then bring total volume to 236.6 mL with water. Store protected from light and cool.

I don’t believe that you want the plant material of the flower sac in contact with the pollen for long term storage.

The main concern is keeping the pollen dry and green material will contain at least some unwanted moisture.
