I think tissue culture is one of those things that I would like to learn but never will unless there is a short seminar on doing it (or maybe do a thread on it) Tissue culture really is super neat. A guy in wash state was telling me that instead of keeping clones under lights, he has hundreds of mothers in his fridge! What a space saver and so much cheaper to keep that way !

What does that mean? Poured 14 tubes? 14 chances for success? Good luck. I would love to see that.

Man, go to that Dropbox link I posted, and pull down the CannaTCInfo.zip file. Inside it there are several books, and one of them is called Plant Tissue Culture, Techniques And Experiments. It’s an introductory textbook on PTC, and, IMO, it’s an excellent resource for basic information.

Yes, for this go around, 14 tubes, means 14 chances. I say tubes, I actually use old baby food jars. There are a couple of places that sell clear plastic caps sized to fit a standard baby food jar. They’re not as narrow as a test tube, so they’re easier to work with.

@Northern_Loki started a thread that has a very detailed guide about the TC process.

I’m in southern Colorado now so I’m back in that same climate. Lots of wild dung around here. I had a bull elk in my back yard a couple of weeks ago. I had heard about one seen around the back of the houses on my road. Then I walk out back one day and had to step over elk shit. Then I did a double take and sure enough I could track where he jumped the fence and pooped his way across my yard to my back door and pooped again and the climbed over the fence to go out of the yard. lol. I wish I’d seen him. We have a lot around here.

Where were you with the dry patties? I used to be a land surveyor in Montana and Wyoming and all over the Rockies and North Dakota. I’ve come across millions of those sun dried cow patties. It is gold for sure.

I was in okanagan county, washington. Beautiful there! I would take a walk everyday for a week or so and fill up 5 gallon buckets. One for horse and one for cow dung. I think somebody could probably make good business selling dehydrated patties! I know I’d be a buyer! Course I always have the option of taking a fresh New York turd and putting it in a solar dehydrator…

First few pics are of a bagseed that made me remember the high school days of 4 fingers for $10. The smell reminded me of what we called Oaxacan. That was a LONG time ago but it was a one of a kind smell , taste. All its own, unforgettable. It may not be tho. ???. It’s been thru 2 frosty nights and several more in the 30’s. The Santa Maria has been out for about a week maybe. I’d have to check the log.

This is very good information.
I never knew one could flip to 12/12 that fast. I “assumed” the plants had to be more mature to start flowering. I used to prune plants to get large kolas, I may of learned a faster better way. But there will be no cloning this way,?, more learning ahead!

I went through that area a long time ago. I was hitchhiking to Montana. I went across the Northern Cascade Mountains to Winthrop, Twisp, etc. It was really pretty on that route.

Hikers paradise through that area. I loved the color of that glacial water in some of those lakes and streams. It just looked cold!

Thats normally for pure long flowering sativas. Flip the lights to 12 12 after first set of true leaves. It’s really really neat that it works for a shorter flowering plant! And to use a crap incandescent bulb until the switch? Even cooler!

Just catching up on the thread. Still a ways to go. Questions for you? 1) Did you “cook this"when it was added into the soil With the blood and bone meal? 2) Did you add much perlite? Generally 50% castings or compost to 50% perlite is added to a soil mix with the poo, blood ,bone ,rice ,etc and the mix is placed in a bag or some dark area to cook for 3 or more weeks. Was something similar done? If not it could have been yr problem a ways back. Compost and cattle poo are great additives but they contain very little air, which roots need as much as water in the soil. Add 50% perlite (small nugget) to 50% castings, compost, or steer ( yes it’s better than cow) manure for adequate oxygen. An alternative manure is from horses. It is up to 66% air and is the " fluffiest” manure. Cattle, sheep, pig , ( manbearpig)are closer to only 10% air. It looks like the lack of air in your mix caused damping off to those original seedlings or as previously mentioned yr uncooked mix did it. My apologies if I repeated someone elses theory or if I stated the obvious.

Dropbox is a cloud service basically. You can store stuff on it, and share it with people online. Just follow this link, and you should see 3 zip files. The CannaTCInfo.zip file has the canna specific docs it, there’s one about setting up a TC lab, and one about general lab procedures. There’s some really good data in there.

I made my first attempt at this mix 3yrs ago. The bull dung was probably fully composted then . when I made the mix I add the rice. Had no prob from mix. But too many problems from my wife. She even told the police I was growing. I don’t smoke ., yet! Didn’t smoke then. I quit smoking 12 yrs ago when I quit growing. Good job. I was growing just for her. Then I moved from n.texas back home to Houston area. Twice. Since I’ve started the mix. Twice some lieing piece of shit told police lies and had me arrested . while in jail they steal everything I own. Truck tools seeds. Mix and everything!. When I got out last time there was 1 bucket of dark soil/ mix looking stuff and a little pile where my compost pile was. I took a chance and used it. Bad mistake. Killed several plants. I bought store compost . spagnum. Bone and blood and Alaska veg and No2o. That’s where I’m at now . New pile composting. Using my homemade lab and fpj mix with cocowater waiting on willows to bloom I do need to start a worm bin. And build bat houses . I’ll try to dig up the few pics I got from that grow( until I pulled cause wife snitched)

Yeah that’s #9 . the lone survived. I germinated 5 more . after I made sure mix was ok. 1 didn’t pop, and I went on a killing spree during the transplanting. #14 survived but didn’t want to grow, so I kicked it out. It’s doing great now. Last nite I put three more in the mix , very carefully, after 1 day in a jar of rain water with a cple drops of lab and h2o2. The seedlings(2) were cracked and just starting to show tap root. (1) had almost 1/4" root. Going to flip in a cple days I hope

I felt strange hitting a like button on that post. I feel for you. My heart got pumping fast reading that. Assholes that did that to you. May karma act swiftly on that sob. I hope yr able to somehow move on from that experience without becoming bitter. Took yr f’in seeds and yr soil too.?Guessing you know who it was that would do such a low thing. When you steal a man’s tools, you take away his ability to make money . It really narrows down the suspect list to someone with limited DNA doesn’t it? I had my years stash stolen from me 3 days before my wedding… by a newer friend who was in my wedding party. I had watered my plants that yr with my dogs and uncles ashes too. All I can say is karma sometimes acts slowly but it does act. No wonder the plants passed on. The compost was tainted by that douchebag! They are looking good this time around! I have a feeling the survivor is a queen.

Sure looks like a she. Nice and stacked. If it is a male, and it smells like you say, then it’s a really nice one. I think it’s a female though😬 i sure hope things flow smooth for you in yr life for a long while now. i hope this strain is able to bring you to your happy place.! Nice weather down there. Those seedlings should be taking off soon ( bag seeds in the ground right?) Have you researched this Santa Maria? It was used to make white widow right?I have seeds of cabeca de negro. Aka Bahia blackhead. It’s a NE Brazilian landrace. Wonder if their is any relation? It was a candidate as a possible white widow parent as well, but it’s unlikely.
